Cancer stem cell isolation and characterization from lung cancer cells

چکیده

Lung cancer is one of the most common cancers in world and a leading cause death. Cancer stem cells (CSCs) are responsible for tumor initiation. CSCs features include self-renewal capacity, differentiation, high invasion-migration, resistance to chemotherapy. characterized by specific surface markers. CD133 widely used as marker many cancers, including lung cancer. CD133-positive an important because they show characteristics CSCs. less sensitive and/or resistant conventional treatments, remaining viable regenerating tumors after treatment. Treatments that can target will be able destroy more effectively, disappearance tumors. Currently, attempts being made develop therapeutics effectively These studies concern isolation characterization In this study, CD133-negative were isolated from H460 cells, large cell model, MACS method. properties investigated tumorSphere formation assay (2% agar plate), hanging drop qPCR positivity was determined 0.3%. It has been observed CSCs-specific properties. gene expression difference between approximately 25-fold. express pluripotency genes (such OCT-4, SOX-2 KLF-4) compared cells.

Isolation and characterization of cancer stem cells from medulloblastoma.

Brain cancer stem cells are a subset of tumor cells present in several types of brain tumor that evade treatment regimens and are responsible for tumor recurrence. Recent reports on several tumors have suggested that Hoechst 33342 dye exclusion is a powerful technique for isolating cancer stem cell-like side population (SP) cells. In the present study, we attempted to isolate the SP cells from ...

Enrichment and characterization of cancer stem cells from a human non-small cell lung cancer cell line.

Tumor cells from the same origin comprise different cell populations. Among them, cancer stem cells (CSCs) have higher tumorigenicity. It is necessary to enrich CSCs to determine an effective way to suppress and eliminate them. In the present study, using the non-adhesive culture system, tumor spheres were successfully generated from human A549 non-small cell lung cancer (NSCLC) cell line withi...

Isolation and characterization of cancer stem cells in vitro.

The cancer stem cell hypothesis is an appealing concept to account for intratumoral heterogeneity and the observation that systemic metastasis and treatment failure are often associated with the survival of a small number of cancer cells. Whilst in vivo evidence forms the foundation of this concept, in vitro methods and reagents are attractive as they offer opportunities to perform experiments ...

Cancer stem cells: therapeutic targets

Cancer stem cells (CSCs) have been identified as rare cellular populations in many cancers, including leukemia and solid tumors. This minor subpopulation of cancerous cells is immortal tumor-initiating cells which thought to be responsible for cancer initiation, progression, metastasis, recurrence and drug/radiation resistance. Low proliferative rate, high self-renewing capacity, differentiatio...

Lung Cancer Stem Cells

Lung cancer remains a major cause of cancer-related lethality because of high incidence and recurrence in spite of significant advances in staging and therapies. Recent data indicates that stem cells situated throughout the airways may initiate cancer formation. These putative stem cells maintain protumorigenic characteristics including high proliferative capacity, multipotent differentiation, ...
